摘要 双光子显微成像技术是近年发展起来的一种新型非线性光学成像方法,由于其优异的特性已广泛用于细胞生物活细胞、组织的长时间动态三维成像。文中首先介绍了双光子成像的原理和特点,在此基础上总结了双光子显微成像技术的研究热点;其次并分别就如何获得更低的细胞损害、更快的图像获取速度和更高的成像灵敏度三个方面作了论述;最后展望了双光子显微成像技术的发展前景。

Abstract: Two-photon fluorescence microscopy technology as a new kind of advanced nonlinear imaging approaches is wildly used in the field of live cell or tissue imaging,especially, which is focused on dynamic three-dimensional cell imaging in a long period. In this paper, the principle and characteristic of two-photon fluorescence microscopy are presented in first. Then, the study focuses of two-photon imaging technology in the world for scientists are pointed out. And, three key aspects of the viability of the specimen, sensitivity of detection, and speed of acquisition are discussed in detail. In the end, the future prospect of development and application of two-phonon imaging technology are predicted
